Abstract

Preparation of nano silver was done by chemical reduction method from precursor AgNO3 and Curcuma domestica as a bioreductor. In this the Nano silver preparation process was carried out with varied of AgNO3 concentration and type of reducing agent. Extraction of Curcuma domestica Val was performed using digestion maceration with 96 % ethanol. Samples were divided into two samples i.e. sample A (with the addition of 1 % PVP) and sample B (Without the addition of 1 % PVP), the second sample was reduced for 7 days using 10 mM AgNO3. Silver Nano particles was detected through by the changing of the color, pH, UV-Vis spectrophotometer measurements, FT IR, SEM, and PSA. The result showed that the UV-Vis spectrophotometer measurements of sample A has λ max 423 nm and sample B has not obtained sprectum λ max. Measurement of FT IR (Fourier Transform Infrared Spectroscopy), obtained the alcohol functional group, alkane group, an aromatic ring group, and the group of alkanes in sample A and ethanol extract of turmeric (Curcuma domestica Val.) sample. Measurment SEM (Scanning Electron Microscopy) obtained the existence of Ag+ ions in the form of small round silver-colored almost shiny, and PSA (Particle Size Analyzer) was obtained by the size of sample A is 279.72 nm. Based on characteristics of the results using UV-Vis Spectrophotometer instrument, FT IR, SEM, and PSA showed that the silver nanoparticles have been formed on the extract of Curcuma domestica. Nano silver formed was tested as antibacterial using disc diffusion method, and the bacteria used were S. aureus. Nano silver concentrations variations are 1%, 2%, 4%, 6%, 8%, and 10% with diffusion agar method. The result showed that the concentration of Nano silver to 10 % as a reducing agent gave higher antibacterial activity than the others up to 10.32 mm.
