Abstract

Antibiotic resistance of microbes has been the main issue worldwide. Thus, discovery of novel antibiotics is very vital particularly antibiotics for antibiotic-resistance microbes. Actinomycetes is a group of filamentous-bacteria that known as the antibiotics producers, better than the other microbes such as bacteria and fungi. The aims research to discover Actinomycetes isolates that have antibacterial activity isolated from several locations in South Sulawesi Province and identify the group of antibacterial compound of selected Actinomycetes isolates. Morphology, physiology, and biochemistry of the selected isolates were evaluated. The characterization involved the spore-chain ornament, colour of mycelium, carbon and nitrogen sources, starch hydrolysis, gelatin hydrolysis, casein hydrolysis, tween hydrolysis, melanin production, resistance on crystal-violet, NaCL, phenol, pH and temperature. Total of 42 Actinomycetes isolates were isolated. By screening using 2 testing microbes, Staphylococcusaureus ATCC 25923 and Escherichiacoli ATCC 35218, total of 3 isolates were able to inhibit S. Aureus ATCC 25923 and total of 1 isolate was able to inhibit E. coli ATCC 35218. KMR 1E2 and SDR 2a showed the highest inhibiting ability towards gram-positive, therefore both KMR 1E2 and SDR 2a were selected for further testing. Bioassay results indicated that etile-extract of the two isolates posse antibacterial activity and methanol-extract of SDR 2a also posses antibacterial activity. Phytochemical results of supernatant extract of KMR 1E2 were to be of alkaloid compound with Rf 0,34. Minimum Inhibitory Concentration (MIC) analysis showed that active compound of KMR 1E2 has MIC on S. aureus ATCC 25923 of 0,3125 mg/ml.
