Abstract The aim of the study was to isolate and characterize the Actinomycetes endophytes and the rizosphere of some of the Fusarium antagonistic onion plantation centers of oxysporum f.sp. cepae and the test capability of the shallot plant (Allium ascalonicum L.) Varieties of Tuktuk. Actinomycetes isolates obtained from the rizosphere and endophytic shallots originating from the five regions namely North Luwu, Soppeng, Enrekang, Gowa, and Bantaeng which further isolates given the name based on the origin of the isolate samples and the origin of the plant sample area onion. Actinomycetes isolates were selected characterised to determine the characteristic morphological, physiological and biochemical characteristics of phenotype. Then test in vitro colonization ability and measurement of photosynthetic pigment levels. The results showed that out of 25 isolates were successfully isolated from the shallot, there were 2 selected isolates representing the origin of the rizosphere and endophyte, namely R2L5 (second rhizosphere isolate from North Luwu of the fifth sample) and E5S5 (fifth endophytic isolate from Soppeng fifth sample) isolates. Based on the antagonistic test it is known that the R2L5 isolates have a 47,41% delay zone and a E5S5 of 33,48%. Results in morphologically characterization of R2L5 isolates have a spherical, white, and the upper surface is convex and does not cling firmly to the media. But gradually grayish and crusty. The E5S5 Isolat has a brownishyellow colony and its surface looks slippery. The results of physiological and biochemical testing showed that R2L5 isolates were able to grow well on 6 types of carbon sources and 2 types of nitorgenous sources, it can grow at 300 and 400C, pH tolerance of 6, 7, and 8. E5S5 isolates are capable of growing well on 4 types of carbon sources and 2 types of nitorgen sources, can grow at temperatures of 300 and 400C, pH tolerance of 6, 7, and 8. The results of photosynthesis pigment pigment content that is chlorophyll a, b, and carotenoid each treatment shows differences. No sterile treatment of endophytic isolates (ETS) have the highest chlorophyll levels.
